Transmission swap
Like a lot of things in modern vehicles, the mechanics of such a swap are not as difficult as the electronics. I suspect you can bolt it in and adapt/kludge the shift mechanism, but getting the electronics to handle it would be the showstopper. Then putting the newer computer in would support the transmission, but other things would be problematic. Not like the pre-computer days when all you had to worry about were the mechanics of a swap.

You are limited to the transmission(s) offered from the factory, and since Dodge offers only one transmission for a given engine and year, there are no alternatives to the stock transmission. If you like the idea of swapping transmissions you need to get a vehicle where multiple transmissions - usually stick vs automatic - are offered, and even then you'd still have to get the correct factory wiring, linkages, pedals, and computer software.
If you want to customize, there are still several things you can change; air intake, throttle body, add blower/turbo, exhaust, tires/wheels, etc. Many of these require computer mods using aftermarket tuners. And almost all will void your warranty. Don't mess around with colder spark plugs without thoroughly researching it.

And the techs have to know all this stuff. Just remember that when you take the car in for service and the shop rate is $110/hour. That's what you're paying for.
Just buy a manual challenger and cut the back half off and weld it to the front half of your Durango. It would be easier than swapping a manual in. Look up manual swaps in cars that actually were available with a manual ( camaros and the like) and you will change your mind. You will need custom driveshaft, flex plate, Master cylinder, slave cylinder,Transmission, Clutch and related parts (flywheel, bearings, etc), Clutch lines
Clutch pedal set, starter with a shorter shaft, custom center console and pray to God that the shifter lines up otherwise you get knuckle buster into the Hvac and a one of a kind tune since your pcm will be commanding shifts that never take place and no info being relayed from the auto trans will likely result in limp home mode or electronic throttle refusing to open or TQ Management that goes on the fritz since it's not getting any transferred torque feedback and only allows 15 lb-ft of torque from idle to redline.
