Default P203 code

I know the code means open circuit on cylinder 3 fuel injector, I just need some guidance on how to test before spending money on parts. As far as actually doing the testing, I have a multi-meter of course, and a test light and continuity tester

I at least confirmed there's continuity between the two posts on the injector itself. Seems like I have to have the engine on to test the power/ground signal of the plug. I had to take the manifold off to get to where I can reach it, but I'm not entirely clear on how to test it. Is that ok to run the engine with the manifold off? I put a few layers of paper towels over the intake, I assume I should take that off before starting the engine? I'm assuming I would either use the test light or the multi-meter for testing the wiring, but I'm not quite clear on how.

Well I just went ahead and replaced the injector without resolving the how-to-test question first and the code is gone now.
