Just had this code come up about a minute ago on start up. #3 cylindar misfire. I felt nothing out of normal, no stumble, hesitation, or anything. I cleared it and it hasnt come back.
Should i worry?, i was thinking it was some cross talk between wires (im getting new ones again). Wires are routed per tsb.

Technically, a 2000-2001 model year Ram shouldn't need the TSB wire routing, but glad you already have that done to eliminate that as a possible solution. I apologize, its been a while since we discussed your upgrades... what wires are you running now?
Could have been crossfire. If you are getting new wires anyway, check (or replace) your distributor cap and rotor as well. I wouldn't worry about it unless it happens again, personally.
