Do you have to remove the drive shaft to to do 4x4 t/case service

There is a clunking sound when in 4x4 going from reverse to drive

Normal. There is slop in the gears, chain, etc, all the way down the driveline.

With the scant info provided by the op in regards to the vehicle with the issue, I would have to agree, it's probably completely normal.
