I get an ignition coil or cylinder misfire every so often with the Superchips tune as well. I only run the performance tune, but the 87 will still cause it under certain conditions.
I wouldn't even reset the truck to stock at first, just clear the code, snipes, odds are it won't come back. If it does, then reset to stock. You could also check the coil pack over that cylinder, they can walk a tad loose on you. Pop it off and back on, just to be sure its tight...

That could very well be it. I would return to stock, clear your DTS codes and try it like that. Every once in a while I will flip that code and its usually vacuum pressure on the coil. If it pops back up in stock, then I would shart checking plugs(and caps) coils ensure the wires aren't pinched etc.
