There is a difference in MPG, but with an SUV this big why drive a V6 or be concerned with fuel mileage? That's what's so great about the Durango, it's a mid size SUV with optional V8! lol
It can tow, it can haul payload, you can enjoy more spirited driving, etc. Me personally, I would not want to get a V6 and every time I saw an R/T think to myself "Dang, what if?" And then proceed to get smoked from a stand still. lol
As far as buying used goes, get an extended warranty if they offer it.

Our highway mileage is consistently 22-24, and I've recorded 26.5 on a loong highway trip to an Ikea (which I'll admit, is a rarity). The return trip, loaded down with at least 1500lb of cargo and three people, was around 21-22.
Unless you're planning on towing something big, I would opt for the V6. I know its a gearhead faux pas to go against the MOAR POWAH mantra, but the V6 really is good enough.

I have a 2012 AWD with the V6. It does what it needs to do and tows a 16' 50 horse alumnacraft all day long without a problem.
However I still regret not getting the v8. The mpg isn't great, but I would give up a little for some more acceleration. Maybe with the new tranny the v6 is enough, but with the 2011-2013 models, I wouldn't do the 6 unless you plan to drive like grandma.

Yes, I test drove the v-6 before the Hemi and thought it was great. But then I test drove the R/T and it was all over.
I am averaging right at 17.3 mpg after 6100 miles and 95% of my miles are at 70 mph, so yeah there is a big difference.
