The search function is a powerful ally!!!
This question comes up about 20x a day forum-wide EVERY TIME gas goes up. Use the search in this section alone and you'll have enough reading material to keep you busy for a whole weekend.
Oh, and brace yourself for the "it's a truck, a 350 HP truck" responses that are inevitable...

Yeah I'm still searching and going thru everything. I not too familiar with Dodge so I'm not sure what's worth looking at replacing/upgrading and what's not and what will give me the most bang for my buck. Understand it's a truck but there's always room for improvement, why waste money if I don't have too.

Make sure the air filter is clean and the fluids reasonably fresh. Some say that a full set of synthetic fluids will provide some benefit, but I'd think it difficult to offset the increased cost. Clean the throttle body, clean/replace the PCV valve, replace the plugs if they have >40K. Plug wires might be in order.
I recently took mine on a road trip after changing the plug wires at 111K. Averaged 78 mph for ~250 miles and got 14.4 mpg. Ignore the computer's report of mileage and calculate it by hand. There is only one way to accurately determine fuel mileage.

the single best thing you can do is your basic tuneup, make sure everything is in working order. Make sure your alignment is good, your fluids are fresh, plugs new, injectors working properly, filters clean, etc. After that you might get some improvements from intake/exhaust work and a programmer to tidy up your A/F ratio. Doing my full headers/exhaust gained me a little mileage but trust me when you feel the kind of power you start building around 3k, and the sound, your foot will be on it.
